BAC Library Vectors Used

The BAC clone libraries SymBio distributes are cloned into one of these vectors:

pBACe3.6 map and information at CHORI, Sequence and features
pTARBAC2.1 map and information at CHORI , Sequence and features
pcc1BAC information from supplier (pdf)

Major pBACe3.6 and pTARBAC2.1 Vector Characteristics:

1. loxP site
2. sacBII gene allows for positive selection.
3. pUC plasmid region disrupts the sacBII gene and allows for high copy number.
4. 6 sites can be used: SacI, SacII, MluI, EcoRI, BamHI and AvaIII.
5. Chloramphenicol resistance (grow at 20ug/ml)
6. Ampicillin resistance (grow at 100ug/ml)
7. T7 & SP6 primer sites
8. Not I sites for cutting out the insert
