Ordering BAC Libraries

Three types of products can be ordered.  Charges are at cost; no profit is derived from this service.

(1) Plates of glycerol stocks of clones
Libraries are stored in this form. On ordering, we create a duplicate of all or part of the complete library, which is then shipped frozen on dry ice and should be stored at -80° C. The number of plates needed for x-fold coverage of each library is shown in the table here. This is most useful if you intent to use these clones for fingerprint mapping or end sequencing or intend to stamp filters for hybridization.  Charges are based on the number of plates ordered.

(2) High Density Filters for Hybridization
Each filter (approximately 8" square) is a positively charged nylon membrane from GENETIX which holds 18,432 spots, i.e., 48 x 384 clones.  The number of filters needed for x-fold coverage of each library can be calculated from the table here.  The filters can be kept indefinitely if stored in a dry, dark place, and with gentle stripping of the probe, can be reused.  Charges are based on the number of filters ordered.

(3) Individual Clones
This is ordinarily done after a successful hybridization (or other indicator) for the clone of interest. These are shipped as agar stabs, which have a finite life at 4° C. We recommend that you streak this to form single colonies on an LB agar plate with the appropriate antibiotic and prepare a glycerol stock of the clones for long term storage at -80° C. While our libraries have little well-to-well contamination it is recommended that a few colonies be verified before initiating any experiments to ensure the clone is pure.
